Withaferin-A

Withaferin – A is a natural compound isolated from the medicinal plant Withania somnifera, an important ingredient in Indian Ayurvedic medicine, and has been widely studied for its anti-inflammatory properties. Recently, studies characterizing the biological activities of withaferin - A have expanded to include the determination of its anticancer properties. Withaferin - A was initially reported to inhibit cell growth and induce apoptosis in in vitro and in vivo models of breast cancer.

Subsequent studies of the anticancer activity of withaferin - A in colon cancer cells demonstrated its ability to modulate Notch1.

Withaferin – A treatment significantly inhibited Notch1 signalling as measured by downregulation of pathway target genes HES1 and HEY1.

In addition, it downregulated numerous prosurvival pathways such as the Akt/NF-κB/Bcl-2 signaling network and the mTOR pathway components pS6K and p4E-BP1. Finally, withaferin- Aactivated c-Jun N-terminal kinase-mediated apoptosis in colon cancer cells.

By using isogenic cancer cells with/without telomerase, Withaferin – A caused stronger cytotoxicity to ALT cells. It was associated with inhibition of ALT-associated promyelocytic leukemia nuclear bodies, an established marker of ALT.

Comparative analyses of telomerase positive and ALT cells revealed that Withaferin – A caused stronger telomere dysfunction and upregulation of DNA damage response in ALT cells. Molecular computational and experimental analyses revealed that Withaferin – A led to Myc-Mad mediated transcriptional suppression of NBS-1, an MRN complex protein that is an essential component of the ALT mechanism. The results suggest that Withaferin –A could be a new candidate drug for ALT cancers.
